However, several recent studies reported a new and exciting role for R-loops in gene expression regulation by influencing transcription termination and chromatin modification. As mutations in several RNA processing factors have been shown to stabilize R-loop formation, we propose here to investigate the link between R-loops and co-transcriptional alternative splicing AS and to decipher the underlying mechanisms, a hypothesis supported by our preliminary results identifying more than splice junctions affected by decreased R-loops upon overexpression of RNase H1. We will use particular AS events as models to elucidate the molecular mechanisms interconnecting R-loops and AS. Moreover, based on our previous expertise, we plan to study the impact of R-loops on DNA damage-induced AS alteration as a model of physiological regulation of AS by R-loops.

Insulin receptor and its relationship with various forms of insulin resistance Insulin binding to its receptor to , plays an important role in maintaining homeostasis of the body. Insulin receptors are present in all vertebrate cells , which reflects a variety of regulatory processes in which the hormone is involved. Adjusting the alternative splicing Alternative splicing is a process in which a single pre-mRNA is formed of more than one isoform mRNA. Type nascent mRNA isoform is the result of a fairly complex regulatory mechanisms that allow to obtain , for example, tissue- specific splicing pattern or changes in cellular splicing profile at various stages of development. This is possible thanks to the cooperation of a number of elements , among which are: the nucleotide sequence and secondary structure of pre- mRNA splicing factors and additional factors proteinaceous or nonproteinaceous. Alternative splicing is one of the key mechanisms extending the complexity of genetic information and at the same time adaptability of higher eukaryotes. As a result, the broad spectrum of isoforms produced by alternative splicing allows organisms to fine-tune their proteome; however, the functions of the majority of alternatively spliced protein isoforms are largely unknown. Ribosomal protein isoforms are one of the groups for which data are limited. Here we report characterization of an alternatively spliced isoform of the ribosomal uL10 protein, named uL10β. The uL10 protein constitutes the core element of the ribosomal stalk structure within the GTPase associated center, which represents the landing platform for translational GTPases — trGTPases. The stalk plays an important role in the ribosome-dependent stimulation of GTP by trGTPases, which confer unidirectional trajectory for the ribosome, allosterically contributing to the speed and accuracy of translation.

Alternative splicing is a cellular process in which exons from the same gene are joined in different combinations, leading to different, but related, mRNA transcripts. These mRNAs can be translated to produce different proteins with distinct structures and functions — all from a single gene. Alternative Splicing. What I think this process of alternative splicing does is let the human genome be fabulously complex while having only 20, proteins encoded in it. And I know 20, sounds like a large number. But remember, we're talking about over 3 billion base pairs. So really, the process of alternative splicing in which each gene, let's say a gene has seven exons.

Alternative splicing AS is a post-transcriptional process that produces transcript variants, thus leading to transcriptome complexity. Recently, the scope of AS studies has been greatly expanded toward clinical applications owing to the abundance of RNA sequencing data. This review consists of two parts. We first summarize bioinformatic resources that are useful for large-scale cancer-related AS studies. We then highlight the research efforts to utilize AS events for predicting clinical outcomes and planning therapeutic strategies. Computational approaches to interrogate AS events have been reviewed under three categories: 1 databases to provide functional and clinical annotation of AS events, 2 analytical tools to identify cancer-associated AS event, and 3 methods to identify splicing-related DNA variants and splicing-derived neoantigens. We also present the recent progress in exploring the clinical utility of AS under four categories: 1 identification of AS events for cancer prognosis, 2 utilization of AS events in molecular classification of various cancers, 3 regulatory mechanisms of AS underlying drug resistance, and 4 potential use of AS in cancer therapy. This review will be helpful for understanding the biological implications of AS in cancer and facilitate the development of AS markers for cancer prognosis and treatment.

Alternative splicing is an important source of heterogeneity underlying gene expression between individual cells but remains an understudied area due to the paucity of computational tools to analyze splicing dynamics at single-cell resolution. We performed extensive benchmarking of MARVEL against available tools and demonstrated its utility by analyzing multiple publicly available datasets in diverse cell types, including in disease.
